When this binding occurs, the nucleus of the cell increases the synthesis of proteins and upregulates metabolic processes. The actual release of testosterone is governed by luteinizing hormone (LH). LH informs the Leydig cells to secrete androgens, including testosterone. Once it has been released, testosterone is controlled by two of the brain’s key mechanisms, albumin and Sex Hormone-Binding Globulin (SHBG). Albumin is present in a higher concentration than SHBG1, and binds to a higher proportion of total testosterone. However, it is done at a lower affinity than SHBG. Due to this, albumin-bound testosterone can be rapidly split up to allow for androgen receptor binding and interaction. SHBG binds to estrogen, though the affinity for estradiol is low, whereas its affinity for testosterone and DHT (dihydrotestosterone) is very high.
In typical cases, less than 2% of your testosterone concentration is considered to be free (active). This testosterone is sometimes known as Bioavailable Testosterone. At least 50% of your circulating testosterone is bound to SHBG. The rest of the circulating testosterone is albumin-bound. SHBG-bound steroids cannot bind to androgen receptors, which is why even the highest levels of testosterone can be inactive and useless.
